What’s the opportunity?
Parabilis is seeking a highly skilled and motivated biochemist/cell biologist with drug discovery experience to join our DeNovo Discovery team. The successful candidate will guide the development and execution of assays to address critical questions pertaining to cellular uptake and endosomal escape of our therapeutic compounds. This will involve working closely with a cross-functional team of chemists, biologists, and data scientists to design and perform experiments that assess Helicon™ uptake efficiency, cytosolic exposure (CE), and endosomal escape mechanisms.
This person will also support ongoing efforts to identify potential next-generation molecules by performing routine testing to measure cytosolic exposure and cellular uptake.
Key objectives include:
- Identify core assays to measure and optimize CE of native Helicons™ and refine our existing CE-related assays to better align with project needs
- Outline key non-CE measurements that are important for contextualizing CE data and incorporate these assays accordingly
- Inform drug design and our understanding of Helicon™ structure-activity relationships through data-driven insights
- Design cell-based functional assays to quantify compound internalization and subcellular localization using techniques such as cell fractionation, mass spectrometry and confocal microscopy.
What you’ll need to be successful:
- Ph.D. in cell and molecular biology, biochemistry, pharmacology or related field with 3+ years of academic or industry experience. Master's degree with 8+ years of industry experience in drug discovery setting.
- Experience with liquid chromatography-mass spectrometry is strongly preferred
- Demonstrated experience in developing cell-based uptake assays and proximity-dependent activity assays, including but not limited to NanoBiT and NanoBRET, is a plus.
- Experience with biophysical/biochemical assay development such as ITC, DSF, MST, FP, TR-FRET, and structural biology, etc. a plus.
- Experience in oncology and peptide drug discovery strongly preferred
- Self-motivation, ability to work independently, excellent organization skills that enable multitasking and working within timelines is required
- Ability to adapt and exercise flexibility for changing platform needs, workflows, and projects
- Strong interpersonal and communication skills are essential
- Able to work on-site and attend in-person meetings
